[Indications] It is a kinase inhibitor suitable for the treatment of Adult patients with chronic phase, accelerated phase, or blast phase chronic myelogenous leukemia who are resistant or intolerant to previous tyrosine kinase inhibitor therapy or have chromosomes that are resistant to or intolerant to previous tyrosine kinase inhibitor therapy Positive acute lymphoblastic leukemia (+). This indication is based on response rate. There are no useful trials demonstrating improvement in disease-related symptoms or increased survival.
[Usage and Dosage] Take orally once a day with or without food.
[Precautions]
Patients with congestive heart failure should be monitored for signs and symptoms of congestive heart failure and treated when clinically indicated.
Hypertension Monitor high blood pressure and treat it when clinical indications indicate.
Monthly monitor for serum esterase disruption or discontinuation in pancreatitis.
Bleeding Interruption for Severe Bleeding
Fluid Retention Monitor patients for discontinuation of fluid retention.
Arrhythmia Monitor for symptoms of arrhythmia.
Myelosuppression, thrombocytopenia, neutropenia, and anemia may require interruption or dose reduction. Monitor complete cell counts biweekly for 1 month and then monthly and as clinically indicated. Interrupt for &/or thrombocytopenia.
Ensure hydration and correction of high uric acid levels before initiating treatment for tumor lysis syndrome
Impaired wound healing and gastrointestinal perforation may briefly interrupt treatment in patients undergoing major surgery.
Embryofetotoxicity may cause fetal harm. Advise women of the potential risk to the fetus.
[Adverse Reactions] The most common non-hematological adverse reactions (&%) are hypertension, rash, abdominal pain, fatigue, headache, dry skin, constipation, joint pain, nausea, and fever. Hematologic adverse reactions include thrombocytopenia, anemia, neutropenia, lymphopenia, and leukopenia.
